Project Title: (Max 1000 characters)	Quantification of GAGAergic neurons in the DNLL of Seizure-prone rats
Project Description:	Generalized seizures result from abnormal excitation of neurons beginning in one brain area and then spreading across the entire brain. This activity is characteristic of epilepsy. When generalized seizures occur repeatedly over a period of time, injury to various brain areas is probable and the physiology of seizure-induced rats may be affected. This project begins with Long Evans Rats that endure an acquired epilepsy procedure focusing on the auditory pathway. The GABA neurotransmitter is a primary inhibitory chemical that works to inhibit seizure activity during the audio-stimulus used to evoke a seizure response. GABA neurotransmission affects the Inferior Colliculus (IC), a site in the auditory pathway where the seizure is initiated. In the Midbrain, GABAergic neuron levels will be accessed in a specific area, the Dorsal Nucleus of the Lateral Lemniscus (DNLL), an extrinsic source of inhibitory neurotransmitters for the IC. The rats acquire epilepsy using an audiogenic seizure model. The rats were primed on pnd 18 using 120 dB tone pips at10 kHz (8s for 8min) to become seizure prone. The seizure events consisted of exposure to a 120 dB Broadband noise for a total of 3 minutes, or until the rat went into a clonic seizure. The rats were sacrificed and perfused intracardially. The brains were removed and stored in a paraformaldehyde solution at 4 degrees Celsius. The tissue will be cut into 50 µm sections. Then, the sections will be cleaned in 0.1% hydrogen peroxidase and rinsed in phosphate buffered saline (PBS). A 10% solution of normal rabbit serum in PBS will be added to the tissue. Then, the Polyclonal antibody to GABA (made in rabbit), will be applied and the sections will be incubated at room temperature. Afterward the secondary antibody will be applied and later the tissue will be rinsed in PBS. Then, the solution containing avidin-biotin complex (ABC) will be introduced to the tissue to bind to the secondary antibody and then later rinsed in PBS. For visualization purposes, the sections will be exposed to diaminobenzidine tetrahydrochloride (DA). Then the tissue will be rinsed in PBS and mounted onto slides. Once the slides are formed, counting of GABAergic cells within the DNLL will be performed. Cells will be counted at 400x magnification using a Pixelink camera for projection purposes onto a computer screen. The cells will be counted in 100x100 um boxes. Then, data analysis will be performed to analyze differences between the rat groups in terms of GABA-producing cell densities. We expect to observe more GABAergic cells in adolescent AGS-prone rats in comparison to rats that received seizure stimulus noise and normal rats, which would suggest that physiological changes occur early in development.
